Evidence: hKSR-2 also regulates the activity of MEKK3 (another MAP3K family member) in HEK-293T cells. We demonstrate that hKSR-2 is a negative regulator of MEKK3-mediated activation of MAP kinase (specifically ERK and JNK) and NF-kappaB pathways, and concurrently inhibits MEKK3-mediated interleukin-8 production.

We have examined the
regulation by
MEKK3 of
MAPK kinase 7 ( MKK7 ) and MKK6, two novel MAPK kinases specific for JNK and p38, respectively

:
Mixed lineage kinase 3 (MLK3) is a mitogen activated protein kinase kinase kinase (
MAP3K ) that
activates multiple mitogen activated protein kinase (
MAPK ) pathways in response to growth factors, stresses and the pro-inflammatory cytokine, tumor necrosis factor (TNF)

(MAP Kinase Signaling System) :
Furthermore, the IL-12/IL-18-driven IFN-? production and MAPK activation in Mekk3-deficient T cells was not affected suggesting that
MEKK3 may selectively
mediate the TCR induced
MAPK signals for IFN-? production
