Cabrero et al., Curr Drug Targets Inflamm Allergy 2002*
Evidence: activation of PPARa represses p65 transactivation of the human IL-6 promoter. This antagonism occurs in a reciprocal manner, since cotransfection of increasing amounts of p65 led to a dosedependent inhibition of a PPARa-mediated activation of a PPAR response element-driven promoter. Transrepression occurs through direct protein-protein interactions between PPARa and the p65 subunit of NF-kB [28].

(decreases, RELA Activity, PPARA Activity)
Delerive et al., J Biol Chem 1999*
Evidence: Transcriptional interference between PPARalpha and both c-Jun and p65 occurs reciprocally, since c-Jun and p65 also inhibit PPARalpha-mediated activation of a PPAR response element-driven promoter.

:
These results indicate that ROS generation and
NF-kappaB activation are
responsible for the down-regulation of
PPARalpha and may provide a new mechanism by which intracellular lipid accumulation occurs in skeletal muscle cells
Kleemann et al., Blood 2003
:
First,
PPAR-alpha increases IkappaB-alpha expression and thus prevents
p50-NFkappaB translocation to the nucleus ... Our findings identify C/EBP-beta and
p50-NFkappaB as novel
targets for
PPAR-alpha and provide a molecular explanation for the reduction of plasma CRP levels by fibrates
Sethi et al., Redox Rep 2002
:
We propose that oxidation of EPA and its
activation of
PPARalpha and subsequent inhibition of
NF-kappaB is the underlying mechanism for the beneficial effects of fish oil

Furthermore, oxidized EPA inhibited NF-kappaB activation in endothelial cells derived from wild-type mice but had no inhibitory effects on NF-kappaB activation in endothelial cells derived from peroxisome proliferator activated receptor alpha ( PPARalpha ) -deficient mice, indicating that oxidized EPA
requires PPARalpha for its inhibitory effects on
NF-kappaB
Keshamouni et al., Neoplasia (New York, N.Y.) 2005
(Carcinoma, Non-Small-Cell Lung...) :
Furthermore,

Besides regulating lipid metabolism, peroxisome proliferator activated receptor (

Dietary intervention with omega-3 polyunsaturated fatty acids contained in fish oil may be useful for the care of IBD patients, and recent studies have stressed the
role of
PPAR on
NFkappaB activity on the potential beneficial effect of dietary lipids on intestinal function

Lack of
NF-kappaB1 ( p105/p50 )

Moreover,
PPARalpha activation

PPAR-alpha overexpression also
inhibited the doxorubicin induced activity of nuclear factor-kappaB

These findings suggest that
NF-kappaB activation in SAM-P8 heart
